What is ICH Q5D?

What is ICH Q5D?

This document provides guidance on standards for the derivation of human and animal cell lines and microbial cells to be used in biotechnological/biological products. It outlines recommendation for the preparation and characterisation of cell banks to be used for production.

What does Livca stand for?

• another using cells at or beyond the limit of in vitro cell age for production. (LIVCA) • Based on production cells expanded under pilot or commercial scale. • Expansion of cells from the WCB; cells from the MCB could be used with appropriate justification.

What is an end of production cell bank?

An End of Production Cell Bank (EoPCB) is sometimes referred to as a Post-Production Cell Bank (PPCB) or as cells at the limit of in vitro cultivation. These cells are tested to validate the production system. This ensures that the cells are stable and that there are no contamination issues within the system.

What are the regulatory requirements for validating a cell line for vaccine production?

1. production, identification and characterization of the cell substrate; 2. validation of the manufacturing process for removal and/or inactivation of adventitious agents; and 3. testing of the bulk and final product to assure safety.

What is a two tiered cell bank?

A two-tier cell bank system consisting of a MCB and WCB guarantees consistent raw material/cellular substrate supply over the life cycle of a biopharmaceutical drug product and beyond. An additional aspect is the storage of cell banks.

What is in vitro cell age?

In vitro cell age: Measure of time between thaw of the MCB vial(s) to harvest of the production vessel measured by elapsed chronological time, by population doubling level of the cells, or by passage level of the cells when subcultivated by a defined procedure for dilution of the culture.

What is MCB and WCB?

MCB=master cell bank, EOP=end-of-production cell bank, WCB=working cell bank.

How do you make a biologic drug?

Biologic drugs are made using highly complex manufacturing processes that involves living cells….Making Biologics

  1. A piece of DNA is inserted into a living cell—a yeast, bacterial, viral, or mammalian cell.
  2. The piece of DNA instructs the cell to produce a large amount of a specific molecule, usually a protein.

What are CHO cells and what are they used for?

CHO cells, which are a cell line derived from the ovary of the Chinese hamster, meant for use in biological and medical research and commercially in the production of therapeutic proteins.

How do you validate a manufacturing process?

End-to-end validation of production processes is essential in determining product quality because quality cannot always be determined by finished-product inspection. Process validation can be broken down into 3 steps: process design, process qualification, and continued process verification.

What are all the criteria that were checked for the validation of the gene edited cell lines?

Functional information. Biological relevance. No information on nucleotide sequence. Genomic DNA of clonal cell lines is sequenced in high throughput.
